So per you lets say I built openfire server for example.com. Added srv
records on my private dns servers and later I downloaded jitsi, given
internal dns servers so users can login to xmpp using JID in jitsi.
In the above scenario desktop sharing, voice and video chat completely be
routed through openfire server or will it happen between user to user?
Again if this is the case rather than diverting a traffic over the
internet, traffic over intranet would be faster...
Please correct me if I am wrong.

It depends: try resolving your servers dns name from inside your
network. if it delivers a private ip (10.x.x.x|192.168.x.x|…) then
clients will not need to connect to the rest of the internet to talk
to the server (and chat, as chat is allways routed over the server).
Before any kind of jingle connection (usaly voice/video) is used it
needs to be established, jitsi uses ICE (interactive connection
establishment) for that which usualy tries to connect to the private
ips so yes: they will use the LAN for desktop sharing.
